Abstract
Rheumatoid arthritis, autoimmune diseases and autoimmune syndromes belong to a special group of diseases in human pathology that manifest clinical, serological and pathohistological signs of activation of autoreactive lymphocyte clones. Activation of the immune system in these diseases is seen in the appearance of autoantibodies to one or more autoantigens. This usually happens when the immune system loses control of part of the lymphocytes that are specifically autoreactive with the cells of their own organism of functional and tissue antigens, or receptors. Antinuclear antibodies are antibodies that are directed against the structural components of the cell nucleus. The research aimed to determine whether treatment with type II collagen will have an effect on the immune system, haematological and biochemical parameters, including ANA in white laboratory rats of the Wistar strain, from both sexes on the 30th and 60th day after immunization. It turned out that induced rheumatoid arthritis leads to different effects on the individual parameters examined, but has no effect on either antinuclear antibodies. Collagen-induced arthritis caused a reaction in 100% of immunized rats with collagen type-II. As an experimental model was used a Wistar breed white laboratory rats, healthy animals of both sexes at seven weeks of age, bred according to standards for breeding experimental animals. They were divided into four groups; control group (n = 20) male animals, control group (n = 20) female animals, experimental group (n = 30) male animals treated with collagen type-II and experimental group (n = 30) female animals who were also treated with collagen type-II. The analyzes were performed on the 30th and 60th day after the immunization. The strictly prepared and highly purified collagen type-II we apply with syringes and small needles with a defined protocol in the knee joint on the back right foot. We took blood for analysis on the 30th and 60th day after the immunization. To determine ANA we used indirect immunofluorescence technique, which is a screening method. From the trials which were made for the effect of collagen induced arthritis on values of ANA in males and females, on the 30th and 60th day of treatment it can be concluded that there is negative ANA values in all groups and in both sexes of experimental animals. From the obtained ANA results in the female group also shows that there is no presence of ANA, as in male rats, but in the female control group there were two individuals where the presence of RNPC antibodies. It’s generally concluded that autoimmunity tests in animal models and paralleling the clinical picture of rheumatoid arthritis in patients has a large contribution to clarifying many issues related to the disease.
