Abstract

Inflammation involved various genes. Gene polymorphisms are contributing factors in the pathogenesis of inflammation. The promontory region of some matrix metalloproteinase’s (MMP) detected polymorphisms of the DNA (those promontory regions controlled transcription of the gene).
Identification of genetic factors which are of enormous meaning for establishing of different profile of patients who will develop chronic periapical lesion or acute odontogenic infection, as well as, calculation of total influence of polymorphisms in promontory region of the gen for matrix metalloproteinase-1 (MMP-1) in the patients was aim of this study.
Election of the material for this study was on the basis of sated clinical diagnoses, after detail analyses and clinical examination with analysis of X-ray changes. We formed three groups. First group was formed from 120 patients after clinically and radiographic verified chronic periapical lesion. Second group formed 40 patients with acute odontogenic infection (Abscessus periapicalis dentoalveolaris). Third (control) group formed 120 patients without endodontic treated teeth and absents of chronic periapical process and acute odontogenic infection.
Standard isolation of genomic DNA from nuclear cells was made with natruim chloride extraction and following precipitation with ethanol. Amplification of the regions of elected genes was made with polymerase chain reaction (PCR).
Correlation between gender and polymorphism of the gene for MMP-1 shows that there was no statistical significance between gender and polymorphism of the gene for MMP-1 in patients with different clinical diagnosis.
Examined polymorphism of the gen for MMP-1 effect on the development of the inflammation and depends from absents or presence of systematic diseases in patients with different clinical diagnosis.
Polymorphism of the gen for MMP-1 in patients in which subjective symptoms are presents and manifested clinical sign determine expression of inflammatory process.
Distribution of the genotypes in the polymorphism for the gene of MMP-1 between patients with different chronic periapical lesion and control group shows differences. Individuals who are carriers of 2G/2G genotype and individuals with 1G/2G genotype (2G carriers) in polymorphism -1607 1G/2G for the gene of MMP-1 with restriction enzyme Alu, shows affection for developing of inflammatory process with more dramatic clinical symptoms and development of one acute odontogenic infection. 1G allele has a protective role for developing of chronic periapical process and acute odontogenic infection. Polymorphism -1607 1G/2G for the gene MMP-1 with restriction enzyme Alu are risk factor for expression of chronic periapical lesions and acute odontogenic infection.
Examined polymorphism of the gene for MMP-1 correlated with chronic periapical lesions and acute odontogenic infections in Macedonian population. Detection of this genetic polymorphism is relevance for obtaining providential treatment of the patients who are with high risk for inflammation.
